Echuca's Sam Edgar and Logan Orr, alongside Echuca United's Oliver Poole, were named in the 24-man squad, it was announced on Monday.
Meanwhile, Leitchville-Gunbower's Oskar Smartt and Rochester's Charlie Palmer also received call ups for their strong body of work.
Pioneers head coach Danny O'Bree said it was a fantastic feeling to lock in the development team.
“These kids have missed out on so much, with COVID last year. This is something we've wanted to get going for a while,” O'Bree said.
“Not only for talent ID, but to give these kids a chance to play some representative football is going to be a really good experience for them.”
Edgar and Orr have lined up for a handful of games in Echuca's under 16 Goulburn Valley League team this year, while Palmer starred in eight games for Rochester's under 16s, working between ruck and forward.
Poole has featured regularly at senior level for United in the Murray Football League, alongside his under 17 commitments, while Smartt has also made an impact at senior level, making his debut for Leitchville-Gunbower in the Heathcote District league while also continuing his service in the under 18 side.
“They're playing some really good football at the moment and some of them at a young age are getting that senior experience,” O'Bree said.
“A couple of them have some good height and some rawness, and some are players with that bit of X-factor.
“We're trying to find those players that stand out in games and those five have a bit of all that in them.”
The squad will be collectively coached by Mick Faulkhead, Luke Crow, Jeremy Rodi as well as former Echuca United coach Guy Campbell.
O'Bree said the best thing about the coaches was they were ex-Pioneers, and a mix of developing and experienced mentors.
“Getting someone like Campbell and his experience and knowledge is pretty cool for our kids to work under,” O'Bree said.
The Pioneers will play two games against the Murray Bushrangers and GWV Rebels, including a June 27 match in Echuca against the Bushies.

O'Bree said the team had its opportunities to win the round eight match.
“Consistently over the four quarters, our decision making wasn't quite up to standard and gave significant chances to Geelong,” he said.
“They seemed a lot hungrier than our kids. That was the difference, so full credit to Geelong.”
In just his second appearance for Bendigo, Tongala's Harley Reid slotted three goals - including two in 90 seconds in the opening quarter - while Kyabram's Anthony DePasquale was among the major possession-getters with 21.
O'Bree remains confident the playing group can rebound ahead of next round.
“They're a pretty proud group and have some really good training standards, so I'm sure they'll be really keen to get back to some good footy,” he said.
